Significance

Palladium-catalyzed enantioselective α-arylation to form tertiary stereocenters was achieved. The formation of tertiary α-stereocenters is in certain respects more difficult than that of quaternary centers as the tertiary products are enolizable. This reaction allows efficient synthesis of the members of the anti-inflammatory profen drug family, such as ibuprofen and ketoprofen, with high yield and enantioselectivity.
