Purification and Characterization of Urinary Stone Protein

Summary:
The character and role of the urinary stone matrix is poorly understood. Our recent study revealed that the cDNA sequence of osteopontin encodes the urinary stone protein. We now present the results of our further study on osteopontin as well as the identification of another stone matrix protein; calprotectin. Calprotectin was extracted from calcium oxalate stone protein with EDTA. The molecular weight was 41.5 kDa. The expression of calprotectin was observed in distal tubular cells and its surrounding connective tissue like osteopontin. This study shows that both osteopontin and calprotectin in the kidney are involved in urinary stone formation as the stone matrix protein.
