A new approach to polyene macrolactam construction is described, based on an intramolecular Stille vinyl-vinyl coupling strategy, e.g. (10) → (12). The methodology is then applied to a synthesis of the macrolactam unit (17) found in the antitumour antibiotic substance leinamycin (1).
