Significance

Palmerolide A was isolated from Synoicum adareanum and exhibits potent and selective activity against the melanoma cancer cell line UACC-62. This highly convergent synthesis is based upon four key reactions including an olefin ring-closing metathesis of a substrate B containing eight double bonds and the enamide side chain in place to construct the macrocycle.
