Protoplast-derived Clones from Cell Cultures of Digitalis purpurea

Abstract

Protoplasts were obtained with high yield from cells of suspension cultures of Digitalis purpurea by incubation with a mixture of cellulase R 10 Onozuka and macerozyme RIO. They were cultivated in a solidified, conditioned Murashige-Skoog medium with 30 μM kinetin and 0.3 M mannitol. Within 24 hours the protoplasts regenerated walls. These single cells underwent subsequent divisions forming clusters of 30-40 cells within 4 weeks. The protoplast-derived cells, as well as the cells of the suspension cultures used for protoplast preparation, obviously divide by means of a centripetal mechanism, deviating from the centrifugal one typical for cells of higher plants.
