Effect of antioxidant activity of Pyrola japonica on H2O2-induced oxidative stress in HSEC cells

Oxidative stress contributes many pathological conditions and disease. However, antioxidant systems are usually effective in blocking oxidative stress. The purpose of this study was to determine the antioxidant activity of Pyrola japonica Klenze ex Alef. [Ericaceae] (TKM2082-W) water extract in H2O2-treated HSEC cells. Total phenolic and flavonoid contents of TKM2082-W were 59.6 mg/g and 26.1 mg/g respectively. The TKM2082-W was screened for chemical antioxidant activity by radical scavenging such as DPPH scavenging, superoxide anion radical scavenging and ABTS+ scavenging assay at different concentration. Throughout the studies TKM2082-W showed marked antioxidant activity. The pretreatment TKM2082-W also decreases intracellular reactive oxygen species (ROS) levels in H2O2-treated HSEC cells. The results of the present study concluded that the TKM2082-W is a source of natural antioxidants which might be helpful in preventing the progress of various oxidative stress mediated diseases including aging.
