Darstellung der terminalen Deoxynukleotidyl-Transferase im histologischen Schnitt*

Abstract

Terminal deoxynucleotidyl transferase (TdT) is a unique DNA-polymerase which is restricted to a few organs (thymus, bone marrow and blood). Therefore, the enzyme is suited for the role of a tumor marker in all other organs. The immunohistological demonstration, hitherto, was mainly hampered by the lack of potent and specific antibodies and by the readiness of the enzyme to diffuse. Now, these problems are solved. The immunohistological demonstration of TdT can be used to classify acute leukemias and lymphoblastic non-Hodgkin-lymphomas of children and to early recognize relapse situations.

Zusammenfassung

Terminale Deoxynukleotidyl-Transferase (TdT) ist eine aus dem gewöhnlichen Rahmen fallende DNS-Polymerase, deren Auftreten auf Zellen weniger Organe (Thymus, Knochenmark und Blut) beschränkt ist. Damit eignet sich das Enzym als Tumormarker in allen anderen Organen. Die immunhistologische Darstellung war bisher vor allem mangels spezifischer Antikörper und durch die hohe Diffusionsbereitschaft des Enzyms erschwert. Diese Probleme sind inzwischen gelöst worden, so daß der immunhistologische TdT-Nachweis als Methode der Subklassifizierung von akuten Leukämien und lymphoblastischen Non-Hodgkin-Lymphomen des Kindesalters und der Früherkennung von Rezidiven eingesetzt werden kann.
