HPLC method development and validation for simultaneous estimation of Metformin and Piperine

Metformin, an oral hypoglycemic drug, used in the management of type 2 diabetes. It controls blood glucose levels in diabetic patients by lowering levels of both basal and postprandial plasma glucose. However, oral bioavailability of metformin is only 50 – 60%, the rate of absorption is slower than its elimination. Hence, a bioenhancer like piperine can be given along with metformin to increase its bioavailability. Various HPLC methods have been reported for estimation of metformin and piperine, individually. However, there are no reports available for simultaneous estimation of metformin and piperine. The present paper discusses the development and validation of a new HPLC method for estimation of metformin and piperine in single run.

An HPLC analysis was carried out using C18 column and the mobile phase, ACN: 0.01 M phosphate buffer (87.5:12.5) at flow rate of 0.9 mL/min. Detection was carried out at 233nm. The developed method was found to be linear over the concentration range 1 – 6 µg/mL for both metformin and piperine. The mean recoveries were found be 99.13% to 100.66% for metformin and 99.58% to 99.67% for piperine. The method was found to be linear, precise, accurate and robust. Thus, a new HPLC method was successfully developed and validated for simultaneous estimation of metformin and piperine.
