Effects of 15-Deoxy-Δ12, 14-prostaglandin J2 on Nuclear Localization of GATA-3 in the Murine Lung in the Presence of Lipopolysaccharide

Summary

15-Deoxy-Δ12, 14-prostaglandin J2 (15d-PG J2) is a regulator of a nuclear transcriptional factor, peroxisome pro-liferator-activated receptor (PPAR)-y. A previous study has demonstrated that 15d-PG J2 enhanced acute lung injury induced by lipopolysaccharide (LPS) in mice. 15d-PG J2 induced mucin-producing cells in the bronchial epithelium, especially in the presence of LPS. The present study investigated the effects of 15d-PG J2 on the activation of GATA-3 and Signal Transducer and Activator of Transcription (STAT) 6, important transcriptional factors in mucus secretion, in the lung in the presence or absence of LPS. ICR mice were divided into 4 experimental groups that intratracheally received vehicle, lipopolysaccharide (LPS: 125 μg/kg), 15d-PG J2 (1 mg/kg), or 15d-PG J2 + LPS. The nuclear localization of GATA-3 and phosphorylated STAT 6 was evaluated 2 h after the intratracheal administration. 15d-PG J2 enhanced the nuclear localization of GATA-3 in the presence of LPS, whereas the nuclear localization of phosphorylated STAT 6 was not altered in the groups. These results suggest that the enhancing effects of 15d-PG J2 on the production of mucin-producing cells might be related, at least in part, to the activation of GATA-3.

Zusammenfassung

Einfluß von 15-Deoxy-Δ12, 14-prostaglandin J2 auf die nukleare Lage von GATA-3 in der Mäuselunge in Anwesenheit von Lipopolysaccharid

15-Deoxy-Δ12, 14-prostaglandin J2 (15d-PG J2) ist ein Regulator für den nuklearen Transkriptionsfaktor Peroxisom-Pro-liferator-aktivierter Rezeptor (PPAR)-y. Eine vorhergehende Studie hat gezeigt, daß 15d-PG J2 bei Mäusen durch Lipopo-lysacharide (LPS) induzierte Lungenschäden verschlimmert. 15d-PG J2 regte Muzin-produzierende Zellen im Bronchialepithel an, insbesondere in der Gegenwart von LPS. In der vorliegenden Studie wurden die Wirkungen von 15d-PG J2 auf die Aktivierung von GATA-3 und den Signalwandler und Aktivator der Transkription (STAT) 6 untersucht, welche sowohl in Gegenwart wie auch in der Abwesenheit von LPS wichtige Transkriptionsfaktoren für die Schleimsekretion in der Lunge sind. ICR-Mäuse wurden in vier experimentelle Gruppen unterteilt, denen der Träger (LPS, 125 µg/kg), 15d-PG J2 (1 mg/kg) oder 15d-PG J2 + LPS intratracheal verabreicht wurde. Die nukleare Lage vonn GATA-3 und der phosphorylierten STAT 6 wurden 2 h nach der intratrachealen Behandlung beurteilt. 15d-PG J2 erhöhte in Gegenwart von LPS die nukleare Lokalisierung von GATA-3, wohingegen sich die Lokalisierung der phos-phorylisierten STAT 6 in diesen Gruppen nicht änderte.

Diese Ergebnisse deuten darauf hin, daβ die fördernden Wirkungen von 15d-PG J2 auf die Muzin-produzierenden Zellen möglicherweise zumindest teilweise von der Aktivierung von GATA-3 abhängen.
