Study of the effect of hormones, medium and explant on the callugenesis of Foeniculum vulgare Mill

Feoniculum vulgare Mill. is one important medicinal plant of Iran [1]. It can be beneficial to modern pharmacy, aromatic, hygienic and food industries, because it contains valuable components in its essential oil [2]. The aim of this investigation was to optimize the tissue culture of F. vulgare. The seeds of F. vulgare were cultured by hydroponic method and after 2 weeks the explants which were provided from epical meristem, hypocotyle, crown and root were transferred to Murashige and Skoog (MS) [3] with 2, 4 and 8mg/L 2,4-D (2,4- Dichlorophenoxyacetic acid) plus 0.25, 0.5 and 1mg/L BAP (6-Benzylaminopurine) [4]. Calli developed within 3 weeks after transferring. Different 2,4-D plus BAP were tasted to select best observation of callogenesis. The highest frequency of callogenesis was found when 2,4-D and BAP were used 2 or 4mg/L and 0.25, 0.5 or 1mg/L, respectively. In conclusion, the results of this study showed that hypocotyl and epical meristem had the best calli in different combination of 2–4-D and BAP tasted.
